# Kisspeptin-10

## What It Is

Kisspeptin-10 is a truncated fragment of the kisspeptin family of peptides, which are products of the KISS1 gene and act as master regulators of the hypothalamic-pituitary-gonadal (HPG) axis. Kisspeptin signals through the GPR54 (KISS1R) receptor on GnRH neurons to trigger gonadotropin-releasing hormone (GnRH) secretion, which in turn stimulates LH and FSH release from the pituitary. This positions kisspeptin at the apex of reproductive endocrine signaling. Clinical research has demonstrated that kisspeptin-10 administration can safely stimulate LH pulses in healthy volunteers and in patients with hypothalamic amenorrhea, offering a physiological alternative to exogenous GnRH. In reproductive medicine, kisspeptin is being investigated as a trigger for oocyte maturation in IVF protocols, with early clinical data suggesting it may reduce the risk of ovarian hyperstimulation syndrome (OHSS) compared to hCG triggers. A 2025 Phase II trial at Imperial College London used kisspeptin-54 as an IVF trigger and reported comparable oocyte yield with significantly lower OHSS incidence. Beyond fertility, kisspeptin is studied for its effects on sexual behavior and psychosexual processing, with fMRI studies showing it enhances brain responses to sexual stimuli. Kisspeptin is not FDA-approved for any indication and remains investigational.

## Why Researchers Study It

Kisspeptin occupies a uniquely important position as the upstream regulator of the entire reproductive hormone cascade. Its potential to replace hCG triggers in IVF and reduce OHSS risk addresses a significant clinical need. The peptide also opens a novel pharmacological target for conditions involving HPG axis dysfunction, including hypogonadism, delayed puberty, and hypothalamic amenorrhea.
